소아시아의 본토 서쪽 17㎞ 지점의 에게해의 큰 섬 레스보스(그 수도의 이름을 따라 미둘레네 라고도 부른다)의 동안에 있던 주요성읍이다. 바울은 제 3차 전도여행에서 돌아오는 길에 이 항구에서 정박했다. 이곳은 일찍부터 에올리아 식민이 행해진 곳 중 하나이며, 시인 알카이오스, 삽포를 낳은 곳이기도 하다. 미둘레네는 본래 작은 섬이었는데, 레스보스 섬에 접합하여 두 양항을 가진 성읍으로 되었다.
